Pharmacodynamics

It was experimentally shown that the components of the drug modify the activity of the ligand-receptor interaction of endogenous regulators with the corresponding receptors: antibodies to the S-100 protein - to serotonin receptors and sigma-1 receptors; antibodies to TNF-α - to the receptor of TNF-α; antibodies to histamine - to localized in the digestive tract N₄- histamine receptors.

The combination of the three active components allows for a complex effect on the central and peripheral links of the pathogenesis of functional disorders of the intestine, including abdominal pain.

Antibodies to protein S-100 possess a wide range of psychotropic activity, including anxiolytic and antidepressive, antiasthenic and nootropic effects, which is clinically manifested in the elimination of internal stress, anxiety, normalization of a number of visceral functions, including activity of the large intestine. Do not have a sedative effect, do not cause addiction, withdrawal syndrome.

Antibodies to TNF-α have a pronounced anti-inflammatory effect, contribute to the normalization of the balance of pro-inflammatory and anti-inflammatory cytokines.

The combined use of components in the complex preparation contributes to the normalization of the nervous and humoral regulation of intestinal function, reducing visceral hypersensitivity of the receptors of the colon to stretching, ensuring the restoration of impaired gastrointestinal motility; stopping the feeling of bloating and stomach overflow; reduce the severity of pain. The antispasmodic effect of the drug is manifested by the relaxation of smooth muscles and a decrease in the tone of the gastrointestinal wall, a decrease in intraluminal pressure, normalization of stool consistency, its frequency and associated symptoms (including relief of imperative urges, tenesmus, feelings of incomplete bowel emptying, reduction of additional efforts during defecation) .

Pharmacokinetics

The sensitivity of modern physicochemical methods of analysis (gas-liquid chromatography, high-performance liquid chromatography, chromatography-mass spectrometry) does not allow to evaluate the content of ultra-low doses of antibodies in biological fluids, organs and tissues, which makes it impossible to study the pharmacokinetics of the Kolofort preparation.
